BSI’s 2X SYBR Green/Fluorescein qPCR Master Mix is a ready-to-use solution that contains components required for quantitative PCR amplification with Bio-Rad iCycler series of real time PCR systems (iQ®, MyiQTM, iQ5TM). The mix includes Taq DNA polymerase, dNTPs, reaction buffer, MgCl2, KCl, a PCR enhancer/stabilizer, SYBR Green and Fluorescein. The user needs only to add template, the primer set and water to the Master Mix to set up the qPCR reaction. This convenient 2X SYBR Green/Fluorescein qPCR Master Mix reduces the time required to set up qPCR reactions and reduces the possibility of contamination particularly when preparing large numbers of reactions. Fluorescein was included in the mix allowing dynamic well factors collection for Bio-Rad iCycler series (iQ®, MyiQTM, iQ5TM) machines. The optimized master mix produces a reliable and sensitive result for specific quantification of target DNA or cDNA from genomic, plasmid and viral templates.

Apoptosis is related to many diseases and induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain containing adapter molecules and members of the caspase family of proteases. These death signals finally cause the degradation of chromosomal DNA by activated DNase. A mouse DNase that causes DNA fragmentation was identified recently and designated CAD (for caspase activated deoxyribonuclease). The human homologue of mouse CAD was more recently identified by two groups independently and termed CPAN and DFF40. Human DFF45 and its mouse homologue ICAD are the inhibitors of CPAN/DFF40 and CAD, respectively. Upon cleavage of DFF45/ICAD by activated caspase, DFF40/CAD is released and activated and eventually causes the degradation of DNA in the nuclei. Activation of CAD/DFF40, which causes DNA degradation, is the hallmark of apoptotic cell death.

Members in the Bcl-2 family are critical regulators of apoptosis by either inhibiting or promoting cell death. Bcl-2 homology 3 (BH3) domain is a potent death domain. BH3 domain containing pro-apoptotic proteins, including Bad, Bax, Bid, Bik, Hrk, Nip3, and Bim, form a growing subclass of the Bcl-2 family. A novel BH3 domain containing protein was recently identified and designated Bnip3L, Bnip3alpha, and Nix (for Nip3-like protein X) (1-3). Bnip3L/Bnip3alpha/Nix is a homolog of the E1B 19K/Bcl-2 binding and pro-apoptotic protein Bnip3. Overexpression of Bnip3L induces apoptosis (2,3). Bnip3L interacts with and overcomes suppresses by Bcl-2 and Bcl-xL. Bnip3L is localized in mitochondria. The messenger RNA of Bnip3L is ubiquitously expressed in human tissues (1,2). Bnip3L and Bnip3 form a new subfamily of the pro-apoptotic mitochondrial proteins.

In healthy cells, Bmf associates with the dynein light chain 2 (DLC2) component of the myosin V motors and is sequestered by the cell's actin cytoskeleton. Disruption of the actin cytoskeleton, either by depolymerization of actin filaments or by detachment of cells from the extracellular matrix, triggers release and activation of Bmf, initiating the downstream apoptotic program (1,2). Apoptosis is related to many diseases and development. Members in the Bcl-2 family are critical regulators of apoptosis by either inhibiting or promoting cell death. Bcl-2 homology 3 (BH3) domain is a potent death domain. BH3-only proteins, including Bad, Bid, Bik, Hrk, Bim, Noxa, and PUMA, form a growing subclass of the Bcl-2 family. A novel BH3-only protein was recently identified in human and mouse and designated Bmf (for Bcl-2-modifing factor) (1). The BH3 domain in Bmf is required both for binding to Bcl-2 proteins and for triggering apoptosis. In healthy cells, Bmf associates with the dynein light chain 2 (DLC2) component of the myosin V motors and is sequestered by the cell's actin cytoskeleton. Disruption of the actin cytoskeleton, either by depolymerization of actin filaments or by detachment of cells from the extracellular matrix, triggers release and activation of Bmf, initiating the downstream apoptotic program (1,2). Bmf is constitutively expressed in many tissues (1,2).

Adhesion to extracellular matrix regulates cell survival through both integrin engagement and appropriate cell spreading. Anoikis is the molecular mechanism of apop-tosis induced by integrin detachment (1). Bit1 (Bcl-2 inhibitor of transcription 1) was recently identified as being involved in this process (2). Bit1 is a mitochondrial protein that is released into the cytoplasm upon onset of apoptosis where it forms a complex with AES, a small Groucho/transducin-like enhancer of split (TLE) protein and induces caspase-independent apoptosis. Both AES and TLE proteins are transcriptional co-repressors that play important roles in neurogenesis, segmentation, and sex determination (3). It has been suggested that Bit1-AES complexes turn off a survival-promoting gene transcription program controlled by TLE (2). Interestingly, apoptosis of cells transfected with Bit1 and AES could be inhibited if the cells were allowed to attach to fibronectin through the alpha5beta1 integrin suggesting that the Bit1-AES pathway contributing to anoikis is regulated by integrins, and in particular, the alpha5beta1 integrin (2).
