PIG-Y Antibody

Glycosylphosphatidylinositol (GPI) lipid anchoring is an important post-translational modification of proteins that takes place in the endoplasmic reticulum. The synthesis of GPI is initiated by GPI-N-acetylglucosaminyltransferase (GPI-GnT), a complex of proteins including PIG-A, PIG-H, PIG-C, GPI1, and DPM2. PIG-Y, the mammalian homolog to yeast Eri1p, is also thought to be involved in the biosynthesis of GPI. The PIG-Y gene encodes two proteins, one of which arises from leaky scanning of the mRNA. This antibody only detects isoform 1 of PIG-Y. Despite its predicted molecular weight, PIG-Y often migrates at 28-30 kDa in SDS-PAGE.

DC-SIGN Antibody

Dendritic cells (DCs) that control immune responses were recently found to capture and transport HIV from the mucosal area to remote lymph nodes (1), where DCs hand over HIV to CD4+ T lymphocytes. DCs also amplify the amount of virus and extend the duration of viral infectivity. Multiple strains of HIV-1, HIV-2 and SIV bind to DCs via DC-SIGN (2). ICAM-3 is the natural ligand for DC-SIGN (3). A DC-SIGN homologue (termed DC-SIGNR, L-SIGN, and DC-SIGN2) was identified recently (4-8). DC-SIGN forms a novel gene family with DC-SIGNR and many alternatively spliced isoforms of DC-SIGN and DC-SIGNR (8). The expression of DC-SIGN was found in mucosal tissues including placenta, small intestine, and rectum.

TNFRSF14 Antibody

Tumor necrosis factor receptor (TNFR) superfamily members are defined by cysteine-rich domains in their extracellular regions that bind TNF-related ligands that share a common structural homology in their extracellular domain. TNFRSF14 was initially identified as the Herpesvirus entry mediator and upon binding to the herpes simplex virus (HSV) envelope glycoprotein D or either of its natural ligands LIGHT and lymphotoxin alpha (LT), activates the transcription factors NF-kappaB and AP-1. Activation of this signal transduction pathway in T cells stimulates T cell proliferation and cytokine production, leading to inflammation and enhanced CTL-mediated tumor immunity, suggesting that these proteins may be useful as potential targets for controlling cellular immune responses.

GFR alpha 2 Antibody

Members of the glial cell line-derived neurotrophic factor (GDNF) family, including GDNF and neurturin (NTN), play key roles in the control of vertebrate neuron survival and differentiation. Physiological responses to NTN require the presence of a novel glycosylphosphadidylinositol linked protein NTNRalpha, which is a cell surface receptor for NTN. The cDNAs encoding NTNRa from human, rat, chicken, and mouse have been cloned recently. NTNRalpha was also termed GDNFRbeta, Ret ligand 2 (RETL2) or TGFbeta-related neurotrophic factor receptor 2 (TrnR2) and nominated as GFRalpha-2 recently. GFRalpha-2 binds NTN and mediates activation of RET receptor tyrosine kinase by both NTN and GDNF. Thus, NTN, GFRalpha-2, and the Ret PTK form a complex to transduce NTN signal and to mediate NTN function.

IL-21 Antibody

A novel cytokine was recently identified in human and mouse and designated IL-21 , which has significant homology to IL-2, IL-4, and IL-15. The receptor for IL-21 (IL-21R, also termed NILR for novel Interleukin receptor) is a new member of the class I cytokine receptor family (1,2). IL-21R forms a complex with the common cytokine receptor g chain, gc, and mediates IL-21 signaling (3,4). IL-21 and its receptor activate JAK-STAT signaling pathway. IL-21 is expressed in activated T cells, and HL-60 and THP-1 cell lines. IL-21 plays a role in the proliferation and maturation of NK, B and T cell populations.

erbB-2 Antibody

The ErbB family consists of four closely related tyrosine kinase receptors that act as potent mediators of normal cell growth and development. Aberrant expression or function of one or more of these receptors can play a major role in the development and evolution of cancer. ErbB-2, also known as HER2, has been implicated in the evolution of both breast and gastric cancers, and is evident in other cancer types such as ovarian and salivary gland tumors. ErbB-2 possesses an active tyrosine kinase domain, but no direct ligand has been identified yet. ErbB-2 is the preferred binding partner to the other members of the ErbB family and is thought to act primarily through the Ras-MAPK, PI3k-PKB/Akt, and PLC-PKC signaling pathways. Numerous anti-cancer strategies have been employed against erbB-2, such as antibody-based therapies to prevent ligand binding or receptor activation through dimerization, antibody-dependent cell mediated cytotoxicity, in addition to direct kinase inhibition to prevent molecular activation/downstream signaling.
