IL-16 was initially identified as a chemotactic cytokine, but is now known to possess a wide range of activities. Later studies have more fully characterized IL-16 as an immunomodulatory cytokine that contributes to the regulatory process of CD4+ T cell recruitment and activation at sites of inflammation in association with asthma and several autoimmune diseases. The precursor of IL-16 (pro-IL-16) is thought to be cleaved towards the C-terminal region by Caspase-3, releasing a 20 kDa active form that binds to and signals through CD4. Besides acting as a chemotactic cytokine, IL-16 is thought to also be involved in the regulation of T cell proliferation and multiple infectious, immune-mediated, and autoimmune inflammatory disorders including irritable bowel syndrome, systemic lupus erythematosus, and neurodegenerative disorders. At least two isoforms of IL-16 are known to exist; the longer isoform (also known as NIL-16) is detected only in neurons of the cerebellum and hippocampus. This antibody will only detect the NIL-16 isoform.

Type I Interferons (IFN-a/b) are produced primarily in response to viral infection by “Natural IFN-producing cells" (IPCs) as part of the host immune response and can also inhibit the development of tumors. IFN-b binding by its receptor results in the activation of the tyrosine kinases Jak1 and Tyk2 and phosphorylation of members of the STAT family of transcription factors, leading to the transcription and expression of the immune response genes. More recently, several members of the toll-like receptor (TLR) family were found to stimulate the production IFN-b. IFN-b is currently used clinically for treatment of tumors, infections and multiple sclerosis.

EBI3 is a subunit in two distinct heterodimeric cytokines: interleukin-27 (IL-27) and IL-35 (1-3). Like interleukin-23 (IL-23), IL-27 is a recently discovered member of the IL-6/IL-12 family of proinflammatory and immunoregulatory cytokines. It exists as a heterodimer composed of the p40-related protein EBI3 and an IL-12 p35-related protein termed p28. IL-27 is produced after activation by antigen-presenting cells and induces proliferation of naïve but not memory CD4+ T-cells. It acts by binding to its receptor WSX-1 (also known as TCCR) and gp130 which results in the activation of a Jak/STAT signaling cascade, suggesting the IL-27 is involved in the regulation of immune processes. It has been suggested that IL-27 can also be used as a therapeutic agent against cancer as it can also induce tumor-specific anti-tumor activity mediated through CD8+ T-cells, IFN-g, and T-bet. IL-35 is composed of EBI3 and the p35 subunit of IL-12 and has been reported to have therapeutic effects against collagen-induced arthritis by expanding the population of regulatory T cells and suppressing Th17 cells. Nucleotides

RNA, like DNA, is a polymer of nucleotides. Each nucleotide consists of a sugar that is attached to a phosphate group and any one of four bases. The RNA polymerase, as it builds the chain of nucleotides, processes only one of the two complementary strands of DNA. This DNA strand is referred to as the template strand. The least confusing name for the other DNA strand is "the nontemplate strand.

Common RNA

Before the nucleotides are linked together, they exist separately as ribonucleoside triphosphates (NTPs). As shown below, the NTPs contain one of the four common RNA bases, A, C, G, and U, linked to a five-carbon ribose sugar, linked, in turn, to a chain of three phosphate groups. During RNA synthesis, a covalent, "phosphodiester" bond is formed between one of the three phosphate groups on one NTP and a hydroxyl group on another. The two other phosphate groups that were part of the original NTP are released.

RNA Synthesis

RNA synthesis is said to proceed in the 5′ to 3′ direction, reflecting the fact that the attachment of new nucleotides always occurs at the 3′ hydroxyl group of the growing RNA chain. RNA synthesis goes through phases that are typical of polymerization processes: initiation, elongation, and termination, yielding an RNA product of defined size and sequence.

The chemoattractant receptor-homologous molecule expressed on Th2 cells (CRTH2) is a recently identified receptor for the prostaglandin D2 (PGD2) in addition to the classic prostaglandin D receptor. CRTH2 is expressed on Th2 cells and eosinophils and mediates chemotaxis of these cells to PGD2 and is thus thought to be a key receptor mediating eosinophil and Th2 recruitment during allergic responses. However, CRTH2-null mice showed enhanced eosinophil recruitment into the lung consistent with observations that the CRTH2-null mice produced significantly higher amounts of interleukin-5 (IL-5) and IL-3. This suggests that CRTH2 plays a nonredundant role in restricting eosinophilia and allergic response in vivo. At least two different isoforms of CRTH2 are known to exist.

Blimp-1 was initially identified as a zinc finger-containing protein that drives the maturation of B lymphocytes into immunoglobulin-secreting cells (1). Blimp-1 exists as at least two different isoforms.

RNA Hybrid


RNA hybrid in biosynthesis is a double-stranded polynucleic acid in which one strand is DNA and the other strand is the complementary RNA; formed during transcription and during multiplication of oncogenic RNA viruses. One of several forms that can be assumed by a double helix. A-DNA is stable in dehydrated conditions.

The adhesion and degranulation adaptor protein (ADAP) was initially identified as a molecular adapter that couples T cell receptor (TCR) stimulation to the avidity of integrins governing T cell adhesion. TCR stimulation promotes the formation of a multi-protein complex containing CARMA1, MALT1, and BCL-10, which through the association of ADAP, ultimately activates the NF-kappaB family of transcription factors. More recent experiments have shown that ADAP controls optimal T cell proliferation, cytokine production, and expression of the Bcl-2 family member Bcl-x(L), suggesting that ADAP regulates T cell activation by promoting antigen-dependent T cell-antigen presenting cell (APC) activation. At least three isoforms of ADAP are known to exist.

Constrained Nucleotide Nucleic Acid


A Constrained Nucleotide, often referred to as inaccessible RNA, is a modified RNA nucleotide. The ribose moiety of an Constrained Nucleotide nucleotide is modified with an extra bridge connecting the 2' and 4' carbons. ZIP10, also known as Slc39A10, is a widely expressed zinc transporter with nine transmembrane domains. Zinc is an essential ion for cells and plays significant roles in the growth, development, and differentiation. ZIP10 mRNA was found to be significantly decreased in the intestines and kidneys of hypothyroid rats and increased in those of hyperthyroid rats, indicating that ZIP10 is positively regulated by thyroid hormones. ZIP10 mRNA was also found to be upregulated in invasive and metastatic breast cancer and cell lines, suggesting that ZIP10 could serve as a possible marker for the metastatic phenotype and possibly a target for novel treatment strategies. At least three isoforms of ZIP10 are known to exist. This antibody will not cross-react with the zinc transporter ZIP11.
