FITC Protein

Bio-Synthesis continued to provide quality life science products and services products and services for the research community, but it has also become a world leader in providing custom peptide products and services. We offer comprehensive services for polyclonal antibody production with large selection for host animals.

FITC Conjugation

Fitc is currently the most commonly-used fluorescent dye for FACS analysis. FITC is a small organic molecule, and is typically conjugated to proteins via primary amines (i.e., lysines). Usually, between 3 and 6 FITC molecules are conjugated to each antibody; higher conjugations can result in solubility problems as well as internal quenching (and reduced brightness). Thus, an antibody will usually be conjugated in several parallel reactions to different amounts of FITC, and the resulting reagents will be compared for brightness (and background stickiness) to choose the optimal conjugation ratio. Fluorescein is typically excited by the 488 nm line of an argon laser, and emission is collected at 530 nm.

FITC Labeled Antibodies

Fluorescein is a fluorochrome that has been widely used in fluorescent assays, and offers the advantage of high quantum yield when attached to protein. However, it has a few disadvantages, including a relatively high rate of photo bleaching, pH sensitivity and a broad-spectrum range that limits its use in multiple labeling. Dyes and R-Phycoerythrin. The choice of fluorochrome to be used is influenced both by the application and questions being asked as well as the available equipment, including filters to measure fluorescent emission wavelengths. The spectral separation of their absorption and emission profiles facilitates the design of multiple labeling experiments.
