Biosynthesis Soil DNA Isolation Kit provides a convenient and rapid method for the detection of microorganisms from soil samples. All types of soil samples can be processed with this kit, including common soil samples and difficult soil samples with high humic acid content such as compost and manure. The kit removes all traces of humic acid using the provided Bead Tubes and a combination of chemical and physical homogenization and lysis. A simple and rapid spin column procedure is then used to further purify the DNA. Total genomic DNA can be isolated and purified from all the various microorganisms found in soil, such as bacteria, fungi and algae. The purified DNA is of the highest quality and is fully compatible with downstream PCR applications, as all humic acid substances and PCR inhibitors are removed during the isolation.

A sample of clay was processed using this Soil DNA Isolation Kit and a leading competitor’s kit. As can be seen, Biosynthesis kit successfully removed more humic acid that the competitor, as
evidenced by the removal of the brown colour in this sample. Humic acids in soil are known PCR inhibitors and inhibit a number of downstream applications.

Total DNA was isolated from samples of clay using this Soil DNA Isolation Kit and a leading competitors kit. Increasing amounts of the purified DNA (1 uL, 3 uL, and 6 uL) were then used as the template in 20 uL PCR reactions using universal prokaryotic 16s rDNA primers. Biosynthesis DNA was successfully amplified in all 3 cases, indicating the high quality of the inhibitor-free DNA, whereas competitor-isolated DNA showed PCR inhibition when 6 uL of DNA was used as the input. Lane M is this HighRanger 1kb DNA Ladder, and the number above each lane indicates the uL of DNA input.

Three different soil samples were processed using this Soil DNA Isolation Kit, and in all 3 cases the humic acid was removed as evidenced by the complete removal of the brown colour in the samples. Tube 1 corresponds to top soil, Tube 2 corresponds to clay and Tube 3 corresponds to potting soil.

Total DNA was isolated from three different soil samples using this Soil DNA Isolation Kit, and the isolated DNA was used as the template in a PCR reaction using universal prokaryotic ITS primers. Bacteria was successfully detected from all 3 samples, including top soil (Lane 1), clay (Lane 2) and potting soil (Lane 3). Lane M is Biosynthesis MidRanger 1kb DNA Ladder.
