This Plasmid MaxiPrep Kit provides a rapid method for preparation of large quantities of high quality, endotoxin-free plasmid DNA. Purification is based on spin column chromatography using this patented resin as the separation matrix. This resin binds DNA under high salt concentrations and releases the bound DNA under low salt and slightly alkali conditions. The purified DNA is free of genomic DNA, RNA and cellular proteins and is suitable for many downstream applications.
This Plasmid MaxiPrep Kit is able to purify up to 1.5 mg of plasmid DNA from 150 mL of bacterial culture containing a high copy number plasmid .

Plasmid DNA from bacterial cultures grown overnight was prepared with the Biosynthesis Plasmid MaxiPrep Kit and compared with two other commercial kits. The yield for each of the triplicates is shown on the graph (B1to B3for Biosynthesis, and Y1 to Y3 and Z1 to Z3 for competitor kits).

DNA isolated with this Plasmid MaxiPrep kit is easily digestible, often requiring less than 1 hour for full digestion. In Figure 2, a 9,309 bp plasmid was purified and digested with Bam HI (lane A), Hind III (lane B), and Sma I (lane c). Lane U is uncut plasmid, and lane M is the Biosynthesis UltraRanger 1kb DNA ladder.
