Human genomic DNA found in saliva can be used in various applications in diagnostics including the detection of biomarkers to diagnose a disease, follow the disease’s progress or monitor the effects of a particular treatment. Isolation of DNA from saliva for diagnostics has become an attractive alternative to isolation from blood or tissue due to the fact that sample collection is non-invasive.
This Saliva DNA Isolation Kit provides a fast and simple procedure for isolating genomic DNA from up to 500 uL of saliva. The Preservation Solution provided with the kit allows for the storage of the saliva samples for extended periods of time at room temperature prior to DNA isolation. Purification is based on spin column chromatography using this proprietary resin as the separation matrix . The genomic DNA is preferentially purified from the other cellular components such as RNA and proteins. The purified saliva DNA is compatible with a number of downstream applications including PCR, Southern Blot analysis, sequencing and microarray analysis.

DNA was isolated from three different 1 mL saliva samples using Biosynthesis Saliva DNA Isolation Kit (Lanes A-C) and run on a 1.8% agarose gel. Note the high yield and quality of the DNA in all lanes. Lane M is Biosynthesis MidRanger 1kb DNA Ladder.

Saliva DNA isolated from 3 different individuals was used as the template in a PCR reaction to amplify the K-ras gene. Lanes A - C contain the expected 157 bp product, and correspond to 3 different saliva DNA samples. Lane D is the negative control and shows only primer dimers, while Lane E is the positive control of 293 HEK DNA, and again shows the expected 157 bp product. Lane M is Biosynthesis FastRunner DNA Ladder.

Hi all,
DNA is isolated from proteins by several methods, including digestion of proteins by the enzyme proteinase K. DNA isolation is a routine procedure to collect DNA for subsequent molecular or forensic analysis. Thanks a lot!
